Artikel des Tages: Classification and basic properties of contrast agents for magnetic resonance imaging

Carlos F. G. C. Geraldes von der Universität Coimbra (Portugal) und Sophie Laurent von der Universität Mons-Hainaut (Belgien) sind zwei Chemieprofessoren mit langjähriger Erfahrung in der Kontrastmittel-Forschung. Aktuell beschäftigen beide sich mit Kontrastmittel für molekulare Bildgebung.

In dem Artikel aus dem Jahre 2008 haben beide alle damals bekannten MR-Kontrastmittel nach Funktionsweise, Anwendung und anderen Kriterien klassifiziert.

"Classification and basic properties of contrast agents for magnetic resonance imaging" ist im Januar 2009 in der Zeitschrift Contrast Media & Molecular Imaging (Volume 4, Issue 1, p. 1-23) erschienen, die ich hiermit auch in meine Linkliste aufnehme.

Artikel des Tages: Slow magic-angle coil spinning: A high-sensitivity and high-resolution NMR strategy for microscopic biological specimens

Der heutige Artikel des Tages widmet sich einer eher technischen Fragestellung.
Erschienen ist er im Februar 2010 in der Zeitschrift Magnetic Resonance in Medicine, Vol. 63, Teil 2, Seiten 269-274.

High-resolution magic-angle spinning has become a valuable tool to study biologic samples; however, there are only a few high-resolution magic-angle spinning methodologies that have been explored specifically to deal with small semisolid biomaterials such as tissues. Here, we report a novel high-resolution 1H NMR spectroscopic approach using magic-angle coil spinning. We demonstrate its potential for intact tissues by combining a resonant microcoil with a large-volume commercial magic-angle spinning rotor, where the microcoil offers incredible sensitivity and the large-volume magic-angle spinning rotor provides stable slow magic-angle spinning. This approach provides high-resolution spectra with high sensitivity and the preservation of living biologic specimens for an accurate chemical analysis.

Artikel des Tages: Protein polymer MRI contrast agents: Longitudinal analysis of biomaterials in vivo

Wir bleiben bei den Polymeren, widmen uns heute aber einer Anwendung in der Medizin.

Abstract:
Despite recent advances in tissue engineering to regenerate biological function by combining cells with material supports, development is hindered by inadequate techniques for characterizing biomaterials in vivo. Magnetic resonance imaging is a tomographic technique with high temporal and spatial resolution and represents an excellent imaging modality for longitudinal noninvasive assessment of biomaterials in vivo. To distinguish biomaterials from surrounding tissues for magnetic resonance imaging, protein polymer contrast agents were developed and incorporated into hydrogels. In vitro and in vivo images of protein polymer hydrogels, with and without covalently incorporated protein polymer contrast agents, were acquired by magnetic resonance imaging. T1 values of the labeled gels were consistently lower when protein polymer contrast agents were included. As a result, the protein polymer contrast agent hydrogels facilitated fate tracking, quantification of degradation, and detection of immune response in vivo. For the duration of the in vivo study, the protein polymer contrast agent-containing hydrogels could be distinguished from adjacent tissues and from the foreign body response surrounding the gels. The hydrogels containing protein polymer contrast agent have a contrast-to-noise ratio 2-fold greater than hydrogels without protein polymer contrast agent. In the absence of the protein polymer contrast agent, hydrogels cannot be distinguished by the end of the gel lifetime.

Artikel des Tages: Heteronuclear chemical shift correlation and J-resolved MAS NMR spectroscopy of lipid membranes

Nach eineinhalb Monaten Weihnachtspause gibt es hier auch endlich wieder einen Artikel des Tages.

Diesmal ist das Thema in der Festkörperphysik zu finden: es geht um NMR-Spektroskopie an DOPC-Membranen, unter der Anwendung der LG-HETCOR-LG-Methode.

Abstract:
Direct observation of J-couplings remains a challenge in high-resolution solid-state NMR. In some cases, it is possible to use Lee–Goldburg (LG) homonuclear decoupling during rare spin observation in MAS NMR correlation spectroscopy of lipid membranes to obtain J-resolved spectra in the direct dimension. In one simple implementation, a wide line separation-type
13C-1H HETCOR can provide high-resolution 1H/13C spectra, which are J-resolved in both dimensions. Coupling constants, 1JCH, obtained from 1H doublets, can be compared with scaled 1JθCH obtained from the 13C multiplets to assess the LG-values efficiency and scaling factor. The use of homonuclear decoupling during proton evolution, LG-HETCOR-LG, can provide J-values, at least in the rare spin dimension, and allows measurements in less mobile membrane environments. The LG-decoupled spectroscopic approach is demonstrated on pure dioleoylphosphatidylcholine (DOPC) membranes and used to investigate lipid mixtures of DOPC/cholesterol and DOPC/cholesterol/sphingomyelin.

Literatursammlung: Einsatz von SPIO in der NMR

Heute möchte ich einiges an Literatur über den Einsatz von superparamagnetischem Eisenoxid (SPIO) in der NMR vorstellen. Das Hauptanwendungsgebiet liegt im medizinischen Bereich und theoretisch könnte SPIO bspw. irgendwann Gd-DTPA und verwandte Kontrastmittel in der kardiovaskulären MRT ablösen.

Der älteste Artikel zum Thema, den ich auf Anhieb gefunden habe, stammt aus dem Jahre 2001 und ist unter dem Titel Quantitative Analysis of SPIO and USPIO Uptake Rate by Macrophages: Effects of Particle Size, Concentration, and Labeling Time in Proceedings of the International Society for Magnetic Resonance in Medicine, Volume 9, p. 880 erschienen.

Die Zeitschrift NMR in Biomedicine hat im November 2004 die Ausgabe 7 von Volume 17 speziell dem Thema eisen-induzierter NMR in biologischem Gewebe gewidmet:
Special Issue: Iron-fortified MRI: effects and applications of iron-induced NMR relaxation in biological tissues

In derselben Zeitschrift ist im vergangenen Jahr ein Artikel erschienen, in dem die Kontrastmittel Gd-DTPA und SPIO bei einer Feldstärke von 3 Tesla verglichen werden:
Measuring SPIO and Gd contrast agent magnetization using 3 T MRI

Abschließend möchte ich noch den Artikel Scaling of transverse nuclear magnetic relaxation due to magnetic nanoparticle aggregation vorstellen, der im Oktober dieses Jahres in der Zeitschrift Journal of Magnetism and Magnetic Materials, Volume 322, Issue 20, erschienen ist.

Artikel des Tages: Visualization of Inert Gas Wash-Out During High- Frequency Oscillatory Ventilation Using Fluorine-19 MRI

Der heutige Artikel beschäftigt sich mit der Lungenbildgebung unter Anwendung von 19F als NMR-Sonde. Die Autoren sind Mitarbeiter des Uniklinikums Mainz bzw. der Universität Mainz.

Aus dem Abstract:
High-frequency oscillatory ventilation is looked upon as a lung-protective ventilation strategy. For a further clarification of the physical processes promoting gas transport, a visualization of gas flow and the distribution of ventilation are of considerable interest. Therefore, fluorine-19 magnetic resonance imaging of the imaging gas octafluorocyclobutane (C4F8) during high-frequency oscillatory ventilation was performed in five healthy pigs. For that, a mutually compatible ventilation-imaging system was set up and transverse images were acquired every 5 sec using FLASH sequences on a 1.5 T scanner. Despite a drop in signal-to-noise ratio after the onset of high-frequency oscillatory ventilation, for each pig, the four experiments could be analyzed. A mean wash-out time (t) at 5 Hz of 52.7 6 18 sec and 125.9 6 39 sec at 10 Hz, respectively, were found for regions of interest including the whole lung.
This is in agreement with the clinical findings, in that washout of respiratory gases is significantly prolonged for increased high-frequency oscillatory ventilation frequencies.
Our study could be a good starting-point for a further optimization of high-frequency oscillatory ventilation.

Artikel des Tages: A dynamic elastic model for segmentation and tracking of the heart in MR image sequences

Heute mal wieder ein Artikel zur Herzbildgebung.

Die Autoren Joël Schaerer, Christopher Casta, Jérôme Pousin und Patrick Clarysse fassen in ihrem Artikel den bisherigen Kenntnisstand zur Bildanalyse in der Herz-MRT zusammen und stellen eine neue Methodik für die Segmentierung und Bewegungsverfolgung bei Herzuntersuchungen vor. Dabei greifen sie die Methode der deformierbaren elastischen Vorlage (deformable elastic template) zurück, die sie um eine zeitliche Dimension erweitern. Dadurch soll das Modell robuster und eine gleichzeitige Auswertung aller zeitlichen Frames möglich werden.

Aus dem Abstract:
Strong prior models are a prerequisite for reliable spatio-temporal cardiac image analysis. While several cardiac models have been presented in the past, many of them are either too complex for their parameters to be estimated on the sole basis of MR Images, or overly simplified. In this paper, we present a novel dynamic model, based on the equation of dynamics for elastic materials and on Fourier filtering. The explicit use of dynamics allows us to enforce periodicity and temporal smoothness constraints. We propose an algorithm to solve the continuous dynamical problem associated to numerically adapting the model to the image sequence. Using a simple 1D example, we show how temporal filtering can help removing noise while ensuring the periodicity and smoothness of solutions. The proposed dynamic model is quantitatively evaluated on a database of 15 patients which shows its performance and limitations. Also, the ability of the model to capture cardiac motion is demonstrated on synthetic cardiac sequences. Moreover, existence, uniqueness of the solution and numerical convergence of the algorithm can be demonstrated.

Artikel des Tages: Investigating the predictive value of whole-brain structural MR scans in autism: A pattern classification approach

Der heutige Artikel liegt mir aus persönlichen Gründen besonders am Herzen.

Die AutorInnen stellen in ihrer Veröffentlichung ein Schema zur Früherkennung von Autismus mittels Magnetresonanztomographie vor. In einer Studie wurden diagnostizierte Autisten und zwei Kontrollgruppen mit symptomatisch ähnlichen Störungen (ADHS) bzw. ohne Störungen auf strukturelle Veränderungen im Gehirn untersucht. Dabei ergab sich offenbar eine signifikante Korrelation zwischen den Veränderungen im Gehirn der autistischen Probanden und der Schwere der autistischen Symptomatik.

Erschienen ist der Artikel im Januar 2010 in Volume 49 der Zeitschrift NeuroImage, Issue 1, Seiten 44-56.

Fundstücke: Einsatz von Jod in der Festkörper-NMR

Der Einsatz von Spin-1/2-Kernen wie Wasserstoff (1H) oder Fluor (19F) in der NMR ist gut bekannt, weniger bekannt ist möglicherweise, daß man auch mit sog. Quadrupolkernen NMR machen kann. Ein solcher Kern ist bspw. Jod, genauer gesagt 127I mit einem Spin von 5/2. Bei einem Kern mit Spin 1/2 ergeben sich aus der Zeeman-Aufspaltung im Magnetfeld zwei Energieniveaus. Allgemein ist die Anzahl der Energieniveaus bei der Zeeman-Aufspaltung proportional zum Spin. Ein Kern mit Spin 3/2 spaltet in 4 Energieniveaus auf, ein Kern mit Spin 5/2 in 6 Niveaus usw., sodaß weitere Übergänge zwischen den einzelnen Niveaus möglich sind.
Man spricht dann z.T. eher von Nuclear Quadrupol Resonance (NQR).

Es gibt einige Webseiten, die über den Einsatz von Jod in der NMR informieren und Literatur auflisten. Vorstellen möchte ich die Seite von Pascal P. Man aus Paris, der eine Menge Infos zu dem Thema zusammengetragen hat und dieses auch einzeln nach NQR-Kernen (z.B. 2D oder 127I) sortiert hat. Sein Informationsangebot ist meines Erachtens gegenüber den anderen Seiten herausragend und ich hoffe, daß er es noch lange pflegt.

Artikel des Tages: An electromagnetic reverse method of coil sensitivity mapping for parallel MRI – Theoretical framework

Es geht wieder weiter - mit einem Artikel über Algorithmen für parallele Bildgebung.

Die Autoren Jin Jin, Feng Liu, Ewald Weber, Yu Li und Stuart Crozier von der University of Queensland in Brisbane (Australien) schreiben im Abstract:

In this paper, a novel sensitivity mapping method is proposed for the image domain parallel MRI (pMRI) technique. Instead of refining raw sensitivity maps by means of conventional image processing operations such as polynomial fitting, the presented method determines coil sensitivity profiles through an iterative optimization process. During the algorithm implementation the optimization cost function is defined as the difference between the raw sensitivity profile and the desired profile. The minimization is governed by the physics of low-frequency electromagnetic and reciprocity theories. The performance of the method was theoretically investigated and compared with that of a traditional polynomial fitting, against a range of system noise levels. It was found that, the new method produces high-fidelity sensitivity profiles with noise amplitudes, measured as root mean square deviation an order of magnitude less than that of the polynomial fitting method. Using the sensitivity profiles generated by our method, SENSE (sensitivity encoding) reconstructions produce significantly less image artefacts than conventional methods. The successful implementation of this method has far-reaching implications that accurate sensitivity mapping is not only important for parallel reconstruction, but also essential for its transmission analogy, such as Transmit SENSE.
